The DNA large fragment polymerase produced by our company is derived from Bacillus subtilis and is obtained by cutting the first 296 amino acid residues of Bacillus subtilis (Bsu)DNA polymerase Ι. The enzyme retains the 5 '-3'  polymerase activity of Bsu, but lacks the 5 '-3' nucleic acid Exonuclease domain, and the large fragment itself lacks 3 '-5' nucleic acid exonuclease activity, which is an isothermal amplification polymerase with chain replacement activity.


1. It improves the incorporation ability of modified nucleotides.

2. Labeling using random primer method.

3. Using for synthesis of the second strand of cDNA and single dA addition to the 3’ end.


1. Due to the lack of 3 '-5' nucleic acid Exonuclease activity, Bsu DNA polymerase (Large fragment)cannot cut off 3 ' unpaired protruding terminal, so it is not suitable for generating flat terminal.

2. At 25 ℃, BsuDNA polymerase (large fragment )retains 50% activity, which is twice that of the Klenow fragment at the same temperature.
